Involvement of prohibitin upregulation in abrin-triggered apoptosis.

摘要: Abrin (ABR), a protein purified from the seeds of Abrus precatorius, induces apoptosis in various types cancer cells. However, detailed mechanism remains largely uncharacterized. By using cDNA microarray platform, we determined that prohibitin (PHB), tumor suppressor protein, is significantly upregulated ABR-triggered apoptosis. ABR-induced upregulation PHB mediated by stress-activated kinase/c-Jun NH2-terminal kinase (SAPK/JNK) pathway, as demonstrated chemical inhibitors. In addition, ABR induced expression Bax well activation caspase-3 and poly(ADP-ribose) polymerase (PARP) Jurkat T cells, whereas reduction specific RNA interference delayed through proapoptotic genes examined. Moreover, our results also indicated nuclear translocation PHB-p53 complex may play role transcription Bax. Collectively, data show plays apoptosis, which be helpful for development diagnostic or therapeutic agents.
