摘要: The enzyme controlled substrate delivery cultivation technology EnBase® Flo allows a fed-batch-like growth in batch cultures. It has been previously shown that this can be applied small vessels such as micro- and deep well plates also shake flasks. In these scales high cell densities improved protein production for Escherichia coli cultures were demonstrated. This current study aims to evaluate the scalability of glucose release technique pilot scale bioreactors. Throughout all scales, is, plates, 3 L bioreactor 150 L cultivations, was very similar model protein, recombinant alcohol dehydrogenase (ADH) produced with yield soluble form. Moreover, EnBase successfully used starter culture density fed-batch cultivations external feeding. Here feeding pump started after overnight Flo. Final optical reached 120 (corresponding about 40 g L−1 dry weight) expression level ADH obtained. ensures initial under mode without need pump. Because linear limitation it provides optimal robust starting conditions traditional feed-based processes.
