摘要: Adjuvant System AS01 is a liposome-based vaccine adjuvant containing 3-O-desacyl-4'-monophosphoryl lipid A and the saponin QS-21. has been selected for clinical development of several candidate vaccines including RTS,S malaria subunit glycoprotein E varicella zoster (both currently in phase III). Given known immunostimulatory properties MPL QS-21, objective this study was to describe early immune response parameters after immunization with an AS01-adjuvanted identify relationships vaccine-specific adaptive response. Cytokine production innate cell recruitment occurred rapidly transiently at muscle injection site draining lymph node postinjection, consistent rapid drainage components node. The induction Ag-specific Ab T responses dependent on Ag being injected same time or within 24 h AS01, suggesting that events occurring postinjection were required these elevated responses. In node, h, numbers activated Ag-loaded monocytes MHCII(high) dendritic cells higher than alone. However, only appeared efficient necessary direct presentation cells. These data suggest ability improve responses, as demonstrated trials, linked transient stimulation system leading generation high number Ag-presenting
