Effects of dexamethasone and dibutyryl cAMP on stanniocalcin-1 mRNA expression in rat primary Sertoli and Leydig cells.

摘要: In this study, STC1 expression levels were determined at various postnatal developmental stages in rat ovaries and testes. The pattern of was found to be sexually dimorphic. addition we examined the effect exogenous glucocorticoids, dexamethasone (DEX), testosterone, follicle stimulating hormone (FSH) dibutyryl cAMP (dbcAMP) on primary Sertoli cell cultures. Our results showed that DEX (via glucocorticoids receptor) stimulated while dbcAMP inhibited cells. Moreover testosterone FSH treatment had no significant inhibitory consistently demonstrated Leydig human chorinoic gonadotropin (hCG) or treated cells, a reduction accompanied with an induction secretion. actions hCG/dbcAMP mediated by PKA, as can rescued co-treatment H89. Using model, modulation regulated transcriptional level actinomycin D significantly reduced respective effect. addition, both effects abolished cycloheximide co-treatment. These indicated regulation treatments indirect may require prior protein synthesis. We suggested de novo synthesis other protein(s) mRNA involved steady-state treatments. Taken together, study provides first evidence male reproductive system.
