摘要: Single-objective stimulated emission depletion (STED) microscopy has long suffered from resolution anisotropy due to the elongated shape of hollow STED spot along optical axis. In this paper, we place a dielectric mirror behind sample and use spatial light modulators illumination beam so that both reflected incident converge same point interfere with each other. By specially setting reflection property mirror, show 3D isotropic focal can be generated while using only one objective lens. When approach is introduced into microscopy, simulated results demonstrate an axial enhancement 3.65-fold together lateral improvement 1.14-fold compared conventional single-objective reached.
