摘要: Aims:  To investigate the use of confocal Raman microspectroscopy (CRM) for analysis structure, composition and development fully hydrated biofilms. Methods Results: Pseudomonas aeruginosa PAO1 biofilms were cultured in a flow cell minimal nutrient medium (artificial sea water) their was followed up to 3 weeks. The spectroscopic signature biofilm cells extracellular polymeric substances (EPS) differentiated distribution colonies within water channels mapped in-plane -depth. initially amorphous, mainly composed with no detectable amount EPS. They developed rapidly give round cellular core enclosed sheath EPS continued increase spread throughout become dominating feature aged colonies. Colonies liquid morphology – characteristic seeding dispersal process also observed. Conclusions:  This study demonstrated that CRM can be used monitor components undisturbed over time. Significance Impact Study:  Confocal facilitates hydrated, live bacterial as function space time, thus making it suitable technique investigating effects various additives environmental factors on growth.
