摘要: Purpose: MicroRNAs (miRNA) are short noncoding RNAs that can play critical roles in diverse biological processes. They implicated tumorigenesis and function both as tumor suppressors oncogenes. The clinical significance of miRNA expression profiles malignant gliomas remains unclear. Experimental Design: In this study, we examined the levels 365 mature human miRNAs 12 gliomas, including 8 glioblastomas 4 anaplastic astrocytomas, using TaqMan real-time quantitative PCR arrays. A validation study was done to corroborate a subset results, miR-196a, -196b, -21, -15b, by analyzing 92 conventional PCR. We modeled relationship between these survival rate 39 glioblastoma patients Kaplan-Meier method multivariate analysis. Results: Expression astrocytomas suggested 16 were candidate markers associated with progression gliomas. Among them, miR-196a showed most significant difference ( P = 0.0038), miR-196b also having high 0.0371). Both increased relative normal brains study. Furthermore, miR-196 significantly poorer 0.0073). Multivariate analysis an independent predictor overall all 0.021; hazard ratio, 2.81). Conclusions: Our results suggest may role be prognostic glioblastomas. Clin Cancer Res; 16(16); 4289–97. ©2010 AACR.
