Interaction of fluorouracil and interferon in human colon cancer cell lines: cytotoxic and cytokinetic effects.

摘要: Fluorouracil (FUra) is the most active agent in advanced colorectal carcinoma, and this activity can be enhanced by various modulating agents both vitro vivo. To determine whether interferon (IFN) capable of augmenting cytotoxic cytokinetic effects FUra, combinations FUra IFN alpha, -beta, -gamma were tested against 2 human colon cancer cell lines vitro. In a clonogenic assay, alpha at concentrations that produced less than 1 log kill, significantly increased lines. gamma also but unlike only highest tested. Median analysis demonstrated all 3 IFNs exhibited synergy with FUra. Combinations no more effective single IFN. Flow cytometric studies indicated these did not correlate alterations. Only combination beta different from those alone. Incubation or for 24 h resulted modest alterations, they modify These results indicate increasing actions separable any interferons.
