1α,25-Dihydroxyvitamin D3 Induces Sphingomyelin Hydrolysis in HaCaT Cells via Tumor Necrosis Factor α

摘要: Treatment of the human keratinocyte cell line HaCaT with 1alpha, 25-dihydroxyvitamin D3 (1,25-(OH)2D3) resulted in hydrolysis sphingomyelin peak elevations ceramide levels after 2-3 h (Geilen, C. C., Bektas, M., Wieder, Th., and Orfanos, E. (1996) FEBS Lett. 378, 88-92). In present paper, mechanism underlying effect 1,25-(OH)2D3 on was investigated. Using culture supernatant cells treated for 2 h, it possible to induce as early 30-60 min addition resting cells. Several lines experimental evidence indicated that tumor necrosis factor alpha (TNFalpha) mediates treatment: (i) stimulated TNFalpha mRNA expression 1 (ii) newly synthesized occurred treatment, (iii) indirect activation by 1, 25-(OH)2D3-treated abolished preincubation antibodies directed against TNFalpha, (iv) neutralizing 55-kDa receptor blocked ability 25-(OH)2D3 These data demonstrate activated an autocrine via expression. Furthermore, this mode action may serve explanation delayed induction vitamin D3.
