摘要: Members of the Plasmodium falciparum var gene family encode clonally variant adhesins, which play an important role in pathogenicity tropical malaria. Here we employ a selective panning protocol to generate isogenic P.falciparum populations with defined adhesive phenotypes for CD36, ICAM-1 and CSA, expressing single distinct variants. This technique has established framework examining expression, its regulation switching. It was found that switching occurs situ. Ubiquitous transcription all variants appears occur early ring stages. However, expression is tightly regulated trophozoites exerted through silencing mechanism. Transcriptional control mutually exclusive parasites express phenotypes. In situ apparently mediated at level transcriptional initiation, as demonstrated by nuclear run-on analyses. Our results suggest epigenetic mechanism(s) involved regulation.
