Quantitative proteomic profiling of the promastigotes and the intracellular amastigotes of Leishmania donovani isolates identifies novel proteins having a role in Leishmania differentiation and intracellular survival.

摘要: Protozoan parasites of the genus Leishmania are important human pathogens that cycle between an extracellular promastigote stage residing in sandflies and intracellular amastigote colonizing phagolysosomal compartment mammalian macrophages. Here, we used isobaric tagging method to quantify global proteomic differences promastigotes amastigotes three different donovani clones derived from THP-1 macrophage cell line. We identified a substantial number differentially modulated proteins involved nutrient acquisition energy metabolism, motility cytoskeleton, transport, signaling stress response. Proteins vesicular trafficking endocytosis like rab7 GTP binding protein, GTP-binding Ras superfamily developmentally regulated protein 1 revealed enhanced expression also putative dynein heavy chain was found be up-regulated it probably has role cargo transport inside vesicles. Significantly, glucose increased eight fifteen-fold, whereas concentrations several associated with cytoskeleton were reduced. Thus, quantitative analysis L. isolates sheds light on some novel may have differentiation survival.
