摘要: Abstract Human peripheral blood monocytes were reproducibly shown to lyse a variety of tumor cells in 3- 4-hr 51 Cr release assay. Ficoll-Hypaque-purified mononuclear suspended medium supplemented with either 10% autologous serum or fetal calf (PCS). With serum, highly purified (97–99%) and viable (>99%) monocyte suspensions obtained by EDTA-reversible adherence plastic surfaces which had been precoated serum. When used as effectors cytotoxicity assays, the recovered from FCS-supplemented exhibited higher cytolytic activity therefore for further studies. Using FCS both coating plates supplementing suspension resulted low activity. Tumor cell lysis measured was detected within 2 hr incubation increased gradually time. The level dependent on effector/target ratio target employed. involvement natural killer lymphocytes observed tumoricidal excluded. Detection cytotoxic short-term assay will be very helpful studies mechanism killing human since potential complicating effects long-term vitro cultivation minimized.
