摘要: Primary cultures of baby mouse kidney epithelial cells can grow without fibroblast overgrowth in a hormone-supplemented serum-free medium (Medium K-1) designed for an established cell line, MDCK. The five supplements Medium K-1 are insulin, transferrin, PGE1, T3, and hydrocortisone. also supports the growth from number animls, including man, withoug overgrowth. Outgrowth explants was observed with K-1. Thus, appears to be selective growth. The physiological properties primary were studied detail. Baby grew at equal rates (0.5 doublings/day) serum-supplemented medium. supported formation colonies low densities. dependence colony upon factors examined. These studies indicated that defined depended all K-1, manner similar, although not identical, MDCK colonies. Primary grown retained cell-associated properties, ability form multicellular domes, phenomenon associated transepithelial salt transport. Amiloride-sensitive Na+ uptake mucosal surface enzyme leucine aminopeptidase cultures. Similar functions monolayers.
