Physical Activity and the Association of Common FTO Gene Variants With Body Mass Index and Obesity

Methods: To determine if variants are associated BMI Old Order Amish (OOA) individuals, to further whether the detrimental associations gene can be lessened by increased activity, a total 704 healthy OOA adults were selected from Heredity Phenotype Intervention (HAPI) Heart Study, investigation geneenvironment interactions cardiovascular disease, for whom objective quantified activity measurements available 92 single-nucleotide polymorphisms (SNPs) genotyped. Results: Twenty-sixFTO SNPs (P=.04 .001), including rs1477196 (P.001) rs1861868 (P.001), 2 moderate linkage disequilibrium (D=0.82; r =0.36). Stratified analyses revealed its association restricted entirely subjects low sexand age-adjusted scores (P.001); contrast, SNP had no effect on aboveaverage (P = .29), genotypephysical interaction achieving statistical significance (P=.01). Similar evidence was also obtained rs1477196. Conclusions: Our results strongly suggest that risk owing susceptibility blunted through activity. These findings emphasize important public health efforts combat obesity, particularly genetically susceptible individuals.
