PD patients also show a slowing of resting state activity. Alpha-synuclein is causally linked to accumulates sleep-related brain regions. While sleep problems over 75% severely impact the quality life caregivers, their study limited by paucity adequate animal models.ObjectiveThe objective this was determine whether overexpression wildtype alpha-synuclein could lead alterations patterns reminiscent those observed measuring sleep/wake activity with rigorous quantitative methods well-characterized genetic mouse model.MethodsAt 10 months age, mice expressing human under Thy-1 promoter (Thy1-aSyn) littermates underwent subcutaneous implantation telemetry device (Data Sciences International) for recording electromyograms (EMG) electroencephalograms (EEG) freely moving animals. Surgeries data collection were performed without knowledge genotype.ResultsThy1-aSyn showed increased non-rapid eye movement during quiescent phase, active wake decreased rapid 24-h period, as well shift density EEG power spectra toward lower frequencies significant decrease gamma wakefulness.ConclusionsAlpha-synuclein produces altered oscillatory PD, model provides novel platform assess mechanisms therapeutic strategies dysfunction PD.
