Estrogen receptor regulation of quinone reductase in breast cancer: implications for estrogen-induced breast tumor growth and therapeutic uses of tamoxifen.

摘要: Antiestrogens have found widespread use in the treatment of breast cancer (reviewed ref. 1). There is also interest tamoxifen as a preventive agent for cancer. However, mechanism antitumor effects antiestrogens relatively unknown. For most part it thought that basis anticancer action inhibition estradiol (E2)-stimulated tumor growth. We observed however can activate detoxifying enzymes, like quinone reductase (NQO1), which protect cells against toxic and tumor-promoting carcinogens (2). Studies characterizing molecular mechanisms behind regulation NQO1 by Estrogen Receptor (ER) support an important role ER pathways regulating antioxidant defenses. Moreover these findings represent novel through function. The activation may contribute to beneficial activity possibly other estrogen target tissues. It possible some induction inhibits genotoxic induced oxidative metabolism estrogens.
