摘要: The last few years have seen a growing interest in the study of DNA methylation because its now acknowledged implication cancer. use bisulfite to convert unmethylated cytosine uracil, even as methylated remains unchanged, constitutes basis for differentiating between and specific CpG sites islands. This technique therefore is critical success this approach. Different parameters be considered order achieve total conversion cytosines uracils. Several bisulfite-based methods are available analyzing status. Methylation-sensitive single-strand conformation analysis (MS-SSCA) yields semiquantitative data. method based on treatment followed by polymerase chain reaction using primers without site avoid selective amplification either or DNA, finally (SSCA). allows one establish clonal variations status clones representing little 5-10% cell population. MS-SSCA has, furthermore, broad application field since it appropriate frozen, fixed, microdissected tissues.
