Hinokitiol inhibits cell growth through induction of S-phase arrest and apoptosis in human colon cancer cells and suppresses tumor growth in a mouse xenograft experiment.

摘要: Hinokitiol (1), a tropolone-related natural compound, induces apoptosis and has anti-inflammatory, antioxidant, antitumor activities. In this study, the inhibitory effects of 1 were investigated on human colon cancer cell growth tumor formation xenograft mice. HCT-116 SW-620 cells derived from cancers found to be similarly susceptible 1, with IC50 values 4.5 4.4 μM, respectively. Compound induced S-phase arrest in cycle progression decreased expression levels cyclin A, E, Cdk2. Conversely, increased p21, Cdk inhibitor. Bcl-2 Bax, cleaved caspase-9 -3. The effect when administered orally was evaluated male BALB/c-nude mice implanted intradermally separately cells. Tumor volumes weights treated (100 mg/kg) both cases. These results suggest that suppression by compound may occur through apoptosis.
