The lac-z reporter gene: A tool for in vitro studies of malignant glioma cell invasion

摘要: A reporter gene (lac-z) was introduced into rat (BT4C) and human (D-54 MG) proliferating glioma cell lines by means of liposomal transfection. Lac-z-transfected cells were first cultured as multicellular spheroids then confronted with fetal brain aggregates. After various intervals the lac-z product, bacterial beta-galactosidase, histochemically detected in cocultures. beta-Galactosidase only which showed an intense blue staining, made them easily distinguishable from tissue. Both a clear pattern migration increasing invasion time tumor infiltrated destroyed Spheroid growth curves no significant differences between transfected nontransfected lines.
