Oxidative stress-induced apoptosis and matrix loss of chondrocytes is inhibited by eicosapentaenoic acid

摘要: Eicosapentaenoic acid (EPA) is an antioxidant and n-3 polyunsaturated fatty that reduces the production of inflammatory cytokines. We evaluated role EPA in chondrocyte apoptosis degeneration. Normal human chondrocytes were treated with sodium nitroprusside (SNP). Expression metalloproteinases (MMPs) was detected by real-time polymerase chain reaction (PCR) apoptosis-related proteins western blotting. Chondrocyte flow cytometry. C57BL/6J mice used for detection MMP expression immunohistochemistry investigation apoptosis. inhibited SNP-induced apoptosis, caspase 3 poly(ADP-ribose) cleavage, phosphorylation p38 MAPK p53, MMP3 MMP13. Intra-articular injection prevented progression osteoarthritis (OA) inhibiting MMP13 treatment can control oxidative stress-induced OA progression, thus may be a new approach therapy.
