摘要: Extracts of 52 TRP3 mutants Neurospora crassa were tested for the presence serologically cross-reacting material by method electrophoretic blot analysis. The test antigen was obtained excision lightly stained bands denatured pure tryptophan synthase after SDS-polyacrylamide gel electrophoresis. Rabbit antisera raised against this neutralized and precipitated native synthase. Of strains, 19 exhibited banding patterns similar to wild type on blots, 25 strains gave no apparent bands, 8 showed unique patterns. This evidence related genetic fine structure mapping data indicate that exhibiting carry missense mutations. Strains which did not exhibit any obvious may have resulted from certain kinds or nonsense mutations frameshift extended deletions. blots interpreted as carrying chain-terminating Genetic place mutant lesions these in a linear order corresponding molecular weights crossreacting protein fragments they exhibit. direction transcription translation locus inferred relationships. organization gene is consistent with ascribed Saccharomyces TRP5 system suggests N-terminal portion corresponds “A” Escherichia coli C-terminal “B” bacterial system.
