摘要: The sphingomyelin pathway is a ubiquitous, evolutionarily conserved signaling system initiated by hydrolysis of the plasma membrane phospholipid to generate ceramide. Ceramide acts as second messenger in activating apoptotic cascade. Diverse cytokine receptors and environmental stresses utilize ceramide signal apoptosis. In several cell systems links stress-activated protein kinase (SAPK)/c-jun (JNK) cascade engagement apoptosis tightly regulated anti-apoptotic control mechanisms, balance between pro- determines magnitude response vitro vivo. This review describes known elements molecular ordering ceramide-mediated mechanisms that regulate its expression. Understanding involved modes their co-ordinated function may yield opportunities for pharmacological interventions with potential clinical applications.
