摘要: In this study, the low molecular weight branched polyethylenimine (PEI) (800 Da PEI) was grafted to biodegradable and biocompatible carboxymethyl dextran (CMD) obtain CMD-g-PEI, plasmid DNA complexed with CMD-g-PEI polycation form polyion complex. The acid-base titration profile showed that had endosomal disruption capacity, agarose gel electrophoresis suggested could condense efficiently. transfection efficiency of CMD-g-PEI/DNA complexes measured by luciferase green fluorescent protein assay in HEK293 cells. results revealed at N/P ratios over 30–70 higher than or comparable 25 kDa PEI optimal ratio (N/P = 10). cytotoxicity as well evaluated NIH3T3 cells, respectively, it also found much lower PEI. resulted high have promising applications when used gene vector. © 2007 Wiley Periodicals, Inc.
