Bioreducible Polypeptide Containing Cell-Penetrating Sequence for Efficient Gene Delivery

摘要: To design excellent polypeptide-based gene vectors and determine the delivery efficiency. Polypeptides (designated as xPolyK6, xPolyK6-R81 xPolyK6-R82), comprising DNA condensing buffering peptide HK6H well cell penetrating (CPP) R8 were obtained by oxidative polymerization of CHK6HC CR8C at different molar ratios in 4 mL phosphate-buffered saline (PBS) containing 30% (v/v) DMSO room temperature for 96 h. The cytotoxicity was studied MTT assay. Moreover, particle size, zeta potential morphology along with vitro transfection efficiency cellular uptake vector/plasmid (pDNA) complexes characterized various w/w to their therapy. All presented ability binding pDNA, additionally low cytotoxicity. Simultaneously, appeared apparent dependence on vector composition. distinct correlation between content demonstrated effective improvement efficacy polymerization. Particularly, xPolyK6-R82 possessed highest a ratio 50. Furthermore, also best capability confocal microscopy flow cytometry.
