Temperature- and phenol-induced alterations in sedimentation rates of 29-S and 18-S ribosomal RNA's from Novikoff hepatoma cells.

摘要: When the ribosomal RNA's of Novikoff rat hepatoma cells (subline NlSl-67) are extracted with hot phenol (60°) plus sodium dodecyl sulfate, 29-S rRNA sediments more slowly, whereas 18-S rapidly than corresponding types cold phenol-sodium sulfate. A similar lowering sedimentation rate is induced by heating phenol-extracted at 50° for 5 min or treatment 95 % dimethyl sulfoxide 7.5 M urea. The change irreversible and accompanied release a 5–6-S RNA. In contrast increase in extraction reversible no other experimental conditions have been found to induce this change. Heating 50, 70 90° urea changes molecule back form that identical rRNA. This not small RNA fragment.
