PMEPA1/TMEPAI knockout impairs tumour growth and lung metastasis in MDA-MB-231 cells without changing monolayer culture cell growth

摘要: Prostate transmembrane protein androgen-induced 1 (PMEPA1)/transmembrane prostate (TMEPAI), a direct target and negative regulator of transforming growth factor beta signalling, has an oncogenic role in many cancers. We observed that knockout (KO) PMEPA1 human breast cancer cell line MDA-MB-231 using CRISPR-Cas9 system resulted reduction vivo tumour lung metastasis but not vitro monolayer capacity these KO lines. This phenomenon was associated with KO-mediated downregulation the key proangiogenic factors vascular endothelial alpha (VEGFA) interleukin-8 (IL8) are essential for growing cells also substantial initiation metastasis.
