An optimal protocol to analyze the rat spinal cord proteome.

摘要: Since the function of spinal cord depends on proteins found there, better defying normal Spinal Cord Proteome is an important and challenging task. Although brain cerebrospinal fluid samples from patients with different central nervous system (CNS) disorders have been studied, a thorough examination specific changes induced by injury or associated to conditions such as neurodegeneration, spasticity neuropathies has yet be performed. In present study, we aimed describe total protein content in healthy rats, employing proteomics tools. Accordingly, developed fast, easy, reproducible sequential protocol for extraction rat cords. We employed conventional two dimensional electrophoresis (2DE) pH ranges (eg. 4–7, 3–11 NL) combined identification mass spectrometry (MALDI-TOF/TOF), well first dimension separation Liquid Chromatography Mass Spectrometry/Mass Spectrometry (LC-MS/MS) maximise benefits this technology. The value these techniques demonstrated here several known neuroglial structures, neurotransmission, cell survival nerve growth system. Furthermore study identified many that not previously described literature which may play role either sensitive biomarkers dysfunction recovery after Injury.
