Separation and analysis of molecular species of mycolic acids in Nocardia and related taxa by gas chromatography mass spectrometry.

摘要: The gas chromatographic separation and mass spectrometric identification of mycolic acids ranging from C28 to C68 in Nocardia related acid-fast groups bacteria was established. Using this method, the molecular species were clearly separated as trimethylsilyl (TMS) derivatives methyl esters, according their total carbon numbers. From ions produced by chromatography spectrometry, double bond numbers determined, while straight branched chain structures identified fragment due C2 ∼ C3 cleavage [RCHOSi(CH3)3]+, C4 [(CH3)3SiOCHCH(R′)COOCH3]+, respectively. Based on composition, average 16 strains taxa (Mycobacterium rhodochrous complex Gordona bronchialis) determined. Each demonstrated possess a characteristic profile acid they shown be classified approximately into at least four groups: (1) C48 (average C33 38) mostly saturated for erythropolis several ‘Mycobacterium complex’; (2) C34 C50 C43 34) monoenoic dienoic N. rubra, corallina lutea; (3) C44 C58 C51 52) tetraenoic asteroides ‘true’ nocardiae; (4) C56 C62 64) pentaenoic polychromogenes bronchialis. results indicate that structural determination spectrometry can one most useful criteria chemotaxonomy bacteria.
