Microarray Analysis of Colorectal Cancer Stromal Tissue Reveals Upregulation of Two Oncogenic miRNA Clusters

摘要: Purpose: Cancer stroma plays an important role in the progression of cancer. Although alterations miRNA expression have been explored various kinds cancers, miRNAs cancer has not detail. Experimental Design: Using a laser microdissection technique, we collected RNA samples specific for epithelium or from 13 colorectal tissues and four normal tissues, microarray gene were carried out. The status was confirmed by reverse transcriptase PCR. Furthermore, investigated whether stromal tissue could influence clinicopathologic factors. Results: Oncogenic miRNAs, including two clusters, miR-17-92a miR-106b-25 cluster, upregulated compared with stroma. Gene profiles cDNA analyses same revealed that putative targets these predicted Target Scan, such as TGFBR2 , SMAD2 BMP family genes, significantly downregulated tissue. Downregulated also found to be involved cytokine interaction cellular adhesion. Importantly, miR-25 miR-92a associated variety Conclusions: highly expressed further validation is required, finding levels factors suggests possibility are crucially progression.
