摘要: The exosome is a protein complex that participates in wide variety of RNA processing, degradation, and quality‐control pathways. conserved all eukaryotes studied to date also present many archaeal organisms, albeit simpler form. To gain insights into the architecture complex, we have chosen hyperthermophilic archaeum Sulfolobus solfataricus as model system. Here describe coexpression, purification, crystal structure determination complexes. understand how binds degrades RNA, designed substrates for degradation experiments exploiting knowledge geometric constraints structure. Furthermore, several structures which were diffused crystals anomalous scattering from 5‐iodo‐uridine–modified was used locate low‐occupancy binding sites.
