Evaluation of minor groove binding probe and Taqman probe PCR assays: Influence of mismatches and template complexity on quantification.

摘要: Real-time PCR assays using 3'-minor groove binder (MGB) or Taqman probes are widely used for clinical virological testing and mutation/polymorphism detection. We compared a 3'-MGB probe to conventional linearity, sensitivity, specificity dynamic range. The performance of the two was plasmids containing different mismatches human genomic DNA as template. Comparable linearity sensitivity were observed MGB assays. Using standard conditions, none sequence-specific. Up five generated detectable signal in assay. well assay influenced by complexity template, latter, however, lesser degree. Overall, these results highlight advantages over regarding mismatch discrimination, but suggest that optimization reaction conditions verification necessary also probes.
