Inhibitory effect of genistein on a tumor promoter-induced c-fos and c-jun expression in mouse skin.

摘要: The effects of genistein, a soybean isoflavone, on 12-O-tetradecanoyl phorbol-13-acetate (TPA)-induced expression c-fos and c-jun in CD-1 mouse skin have been investigated. A promoting dose (8.5 mu mol) TPA significantly increases transcript levels c-fos, 2.7 3.2 kb mRNA by 7.0-, 3.2-, 1.7-fold, respectively. Topical application genistein 30 min before treatment inhibits TPA-induced these protooncogenes. Suppression was more pronounced than that c-jun, at 10 mol almost completely diminished. Genistein exhibited only weak suppressive effect expression. Effect time also results showed topical prior to, simultaneously, after tumor promoter can equally suppress mechanism which proto-oncogene is unknown. However, it appears the inhibition independent protein kinase C (PKC) activation pathway because its PKC activity. It hypothesized inhibitory may be through modulation mitogen-activated (MAP) vivo.
