A dynamic multiple reaction monitoring method for the multiple components quantification of complex traditional Chinese medicine preparations: Niuhuang Shangqing pill as an example.

摘要: It is a challenging task to simultaneously and quantitatively analyze multiple components in DFF [Da-Fu-Fang, namely, complex traditional Chinese medicine (TCM) preparations containing more than ten TCMs] due their numerous extreme chemical compositions possessing wide variety of physical features, very low content. Rather using conventional mass spectrometry (MS) method with reaction monitoring (MRM), the current study, this challenge was addressed by dynamic (DMRM). Using DFF, Niuhuang Shangqing pill, which composed 19 TCMs, as model, rapid (one run 20min), sensitive [lower limit detection (LOD) quantitation (LOQ) were achieved comparable MRM] accessible (a standard HPLC/MS/MS instrumentation employed) MS successfully developed for simultaneous quantification 41 bioactive represented 15 medicinal plants. A comparison LOD LOQ MRM DMRM made reveal that latter demonstrated advantages over former. Meanwhile, operating procedure concerning development new recommended. The data obtained positive ion mode electrospray ionization source, acetonitrile water mobile phase Kinetex C18 core-shell column (100mm×2.10mm, 2.6μm, Phenomenex Inc.) analytical column. This then applied 32 batches samples. transpired, through principal component analysis orthogonal partial least squares discriminant analysis, consistency products relatively good within one company, but poor among different companies samples; failed qualify terms Pharmacopeia. work illustrated proposed particularly suitable quantifying trace capable ensuring quality DFF.
