Machilus thunbergii extract inhibits inflammatory response inlipopolysaccharide-induced RAW264.7 murine macrophages viasuppression of NF-$\kappa$B and p38 MAPK activation

摘要: Machilus thunbergii Sieb. and Zucc. (Lauraceae) is a medicinal plant used to treat edema pain in China Taiwan. The anti-inflammatory effects of M. were not clear. In this study, we evaluated the extract LPS-stimulated RAW 264.7 cells. cells pretreated with various doses (12.5 mg/mL-100 mg/mL) activated LPS. We found that 25 mg/mL inhibited nitric oxide (NO), prostaglandin E2 (PGE2), inducible synthetase (iNOS), cyclooxygenase-2 (COX-2) expression, as well levels proinflammatory cytokines including interleukin (IL)-6 tumor necrosis factor-alpha (TNF-a). Furthermore, also suppressed translocation nuclear transcription factor kappa-B (NF-?B) subunit p65 proteins into nucleus induced dose-dependent inhibition phosphorylation p38 mitogen-activated protein kinase (MAPK). increased HO-1 Nrf2 production concentration-dependent manner. Taken together, these results suggest has an effect because it reduces mediators via suppression NF-?B MAPK activation
