摘要: Abstract Sea cucumber glucocerebrosides (SCGs), an important sphingolipid, have extensive functions in inhibiting the growth of cancer cells, but their potential to promote neuronal outgrowth hasn’t been well studied. Herein, we evaluated neuritogenic effects three Cucumaria frondosa SCGs series (SCG-1, SCG-2, and SCG-3) NGF-induced PC12 cells. These exhibited significant a dose-dependent structure-selective manner, which increased ratio neurite-bearing cells expression axonal (GAP-43) synaptic (synaptophysin) proteins. Mechanistic studies suggested that reinforced phosphorylation TrkA, coupled with ERK1/2 activation, then triggered activation CREB, finally resulted marked increase BDNF expression. Furthermore, could be suppressed when accompanied by additions TrkA inhibitor GW441765 or MEK U0126. In conclusion, possess effect promotes signalling downstream from likely via MEK/ERK1/2/CREB/BDNF pathway.
