Effects of the microtubule stabilizing agent peloruside A on the proteome of HL-60 cells

摘要: Peloruside A, isolated from the marine sponge Mycale hentscheli, has a similar mechanism of action to paclitaxel (Taxol®), drug used clinically treat tumors breast, ovary and lung. Paclitaxel peloruside stabilize polymerized form tubulin arrest cells in G2/M cell cycle. We have therefore two-dimensional electrophoresis proteins examine effect A on human HL-60 promyeloid leukemic line. Our goals included investigation whether affected could be mapped onto pathways that predicted cellular effects this compound. In response 100 nM treatment for 24 h, seventeen identified showed significant change abundance with fourteen increases three decreases. Use Ingenuity Pathways Analysis confirmed consistent known microtubules apoptosis. results also indicate potential role c-Myc actions an induction aneuploidy seen at low concentrations peloruside.
