摘要: Peloruside A (peloruside), a compound isolated from the marine sponge Mycale hentscheli, inhibits growth of human (HL-60) and mouse (32D-ras) myeloid leukemic cells, as well non-transformed 32D cells. Using MTT cell proliferation assay trypan blue dye exclusion tests, little difference was seen in inhibition between 32D-ras cells; however, peloruside more cytotoxic to oncogene-transformed also blocked cells readily G2/M cycle, leading apoptosis. Annexin-V/propidium iodide staining showed that 1.6 μM induced significant death by 36 hours (16% survival), but comparable levels early 14 (11% survival). There no evidence for activation either initiator caspases-8 or -9 0.1 following 12 exposure. inhibited T IL-2 IFN γ production both mixed lymphocyte reaction CD3 cross-linking, this effect shown be non-specific effect. It is concluded preferentially targets over inducing transformed undergo
