Platelet adhesion and arteriolar dilation in the photothrombosis: observation with the rat closed cranial and spinal windows

摘要: The mechanism of cerebral infarction, in which thrombus formation and platelet-endothelium interaction play an important part, have not yet been clearly elucidated vivo. aim this study was to observe rolling adherent platelets analyze leukocytes vessel diameter change vivo using a photothrombotic occlusion model.A photothrombosis, is mediated by free radicals, induced male Wistar rats the presence photosensitizing dye (Photofrin II) exposure filtered light. Rhodamine 6G-labeled were visualized with intravital fluorescence videomicroscopy through closed cranial or spinal window. diameter, photothrombosis leukocyte adhesion analyzed. Rolling observed during irradiation Before recognized, irradiated arteriole dilated significantly. After photochemical arteriole, other arterioles also increased venules. almost completely composed according electron microscopic analysis. arteriolar dilation rate number cerebrum greater than those cord. By combining microscope techniques, we able for first time microvascular responses microvasculature. It suggested that can lead platelet aggregation, role as vasodilator. This model useful microcirculatory analysis investigate interaction, aggregation effect radicals on microcirculation.
