Functional comparison of the transposition core machineries of phage Mu and Haemophilus influenzae Mu-like prophage Hin–Mu reveals interchangeable components

摘要: Bacteriophage Mu uses DNA transposition for propagation and is a model studies in general. Recent identification of Mu-like prophages within bacterial genomes offers new material evolutionary comparative functional studies. One such prophage, Hin–Mu Haemophilus influenzae Rd, was studied its transpositional properties. The components core machinery, the encoded transposase (MuAHin) binding sites, were evaluated properties by sequence comparisons DNase I footprinting. Transpositional activity examined vitro assays directly assessing assembly catalytic function machinery. readily assembled catalytically competent protein–DNA complexes, transpososomes. Thus, encodes contains critical sites. Despite marked differences, machineries are partially interchangeable, reflecting both conservation flexibility functionally important regions transpososome structure.
