摘要: AMONG neural cell lines the C-6 glioma, a line cloned from an N-methylnitrosourea (MNU)-induced experimental glioma in rat, has generated considerable interest as model system for study of neuroglia1. The clone was originally selected on basis high production protein S-100, believed to be characteristic glial cells vivo2. Since its introduction, been further characterised with regard number other biochemical features3. Among these is recent demonstration fibrillary acidic (GFA) by immunofluorescent staining maintained certain vitro conditions4. GFA protein, isolated multiple sclerosis plaques and areas sub-ependymal gliosis central nervous system, shown specific marker astrocytes, component 80–90 A diameter filaments5–8, not reported non-glial lines. It is, therefore, useful differentiation type mature astrocyte. In previous work line, found present immunofluorescence only occasional monolayer or suspension cultures, markedly increased many when they were 12–14 d organ culture using Gelfoam matrices4. Such known favour structural vitro9. No increase filaments observed, however, any systems used, suggesting that synthesis could proceed without formation intracytoplasmic filaments4. We compared quantitative levels types various conditions, “two-site” immunoradiometric assay recently developed this laboratory10.
