Nematicidal activity of three novel extracellular proteases of the nematophagous fungus Monacrosporium sinense

摘要: Extracellular proteases are an important virulence factor for the nematophagous fungi Monacrosporium. The objective of this study was to optimize, purify, partially characterize, and evaluate nematicidal activity produced by fungus Monacrosporium sinense (SF53) solid-state fermentation. Wheat bran used as substrate protease production. variables moisture, pH, incubation time, temperature, glucose, yeast extract, number conidia were tested their influences on production SF53. To determine optimal level selected central composite design applied. crude extract obtained purified in two steps, ion exchange chromatography a gel excision. SDS-PAGE zymogram performed analysis purification process. Proteolytic also at different pHs temperatures. In vitro assay, three evaluated. pH time showed significant effect (p < 0.05) protease. highest value 38.0 (U/ml) under conditions 5.0 211 h. SF53 (Ms1, Ms2, Ms3) which directly from zymogram. Ms1, Ms3 following percentage reduction Panagrellus redivivus compared control after 24 h: 76.8, 68.1, 92.1 %. This is first report use isolate phytonematode, may be research tool future works.
