摘要: The 5' flanking region of the human urokinase (uPA) gene has been fused to reporter chloramphenicol acetyl transferase (CAT) and its activity assayed by transfection in two cell lines. Progressive deletions uPA regulatory from end maintain a high level expression provided at least 1870 (in A1251 cells) or 1963 HFS10 nucleotides are retained. A DNA fragment -2350 -1824 enhancer properties, stimulating transcription an enhancerless SV40 early promoter independently orientation distance. Internal that still retain element reveal presence negative cis-acting sequences between -1572. Their removal, fact, increases transcriptional activity. Differences uPA-CAT fusion genes lines also observed, indicating cell-specific sequences.
