Implementation of buffy‐coat‐derived pooled platelet concentrates for internal quality control of light transmission aggregometry: a proof of concept study

摘要: Essentials In platelet function testing, standardized internal controls (IQC) are not commercially provided. Platelet testing was performed daily on aliquoted pooled concentrates. Pooled concentrates showed stability for control purposes from Monday to Friday. Pooled provide the necessary steadiness serve as IQC material. SummaryBackground Standardized available material quality of light transmission aggregometry (LTA) is still lacking. Moreover, availability normal blood donors fresh platelets difficult in small laboratories, where ‘volunteers’ may be short supply. Objectives To evaluate implementation buffy-coat-derived (PCs) LTA. Methods We used PCs bank LTA. On each weekend one PC prepared (> 200 mL) and original storage bag a basis four baby bags (40–50 mL), which were delivered Friday our laboratory. The measurements at least 85 work-weeks (from Friday) evaluated with this new material. LTA four-channel Chronolog 700 Aggregometer (Chronolog Corporation, Havertown, PA, USA) (agonists: collagen, adenosine diphosphate [ADP], arachidonic acid [AA] thrombin receptor activator peptide-6 [TRAP-6]). Results The medians aggregation ADP AA 68.0–59.5, 3.0–2.0 51.0–50.0%, respectively, mean TRAP-6 71.2–66.4%. Conclusions Buffy-coat-derived reliable robust would beneficial whole laboratory procedure employees’ safety.
