Wavelet Transformations of Tumor Expression Profiles Reveals a Pervasive Genome-Wide Imprinting of Aneuploidy on the Cancer Transcriptome

摘要: Aneuploidy is frequently observed in many human cancers, but its global effects on the cancer transcriptome are controversial. We did a systematic and unbiased genome-wide survey to determine extent tumor9s abnormal karyotype (chromosomal amplifications deletions) detectably “imprinted” onto that gene expression profile. By using novel methodology employing wavelet transform signal-processing algorithms identify genomic regions of coordinated (wavelet variance scanning), we analyzed series gastric cell lines identified >100 exhibiting distinct patterns subtle significant transcription, ranging from tens hundreds genes. A large majority (80%) these could be specifically localized site detectable amplification or deletion; reciprocally, up 47% total aneuploidy each individual directly inferred data. Genome-wide portraits tumor can thus successfully reconstructed solely data, implying must pervasively globally imprinted within transcriptome. may contribute behavior not just by affecting few key oncogenes suppressor genes also subtly altering levels oncogenome.
