摘要: Yarrowia lipolytica LgX64.81 is a non-genetically modified mutant that was previously identified as promising microorganism for extracellular lipase production. In this work, the development of fed-batch process production enzyme in strain described. A lipolytic activity 2,145 U/mL obtained after 32 h batch culture defined medium supplemented with 10 g/L tryptone, an enhancer expression. To maximize volumetric productivity, two different strategies had been investigated. comparison to process, intermittent strategy not improved productivity. contrast, stepwise feeding combined uncoupled cell growth and phases resulted 2-fold increase namely, reached 10,000 80 culture. Furthermore, purified homogeneity by anion exchange chromatography on MonoQ resin followed gel filtration Sephacryl S-100. This overall yield 72% 3.5-fold specific activity. The developed offers great potential economic Lip2 at large scale Y. LgX64.81.
