Adipose-Derived Stem Cells Expressing the Neurogenin-2 Promote Functional Recovery After Spinal Cord Injury in Rat

摘要: Neurogenin2 (Ngn2) is a proneural gene that directs neuronal differentiation of progenitor cells during development. This study aimed to investigate whether the use adipose-derived stem (ADSCs) over-expressing Ngn2 transgene (Ngn2–ADSCs) could display characteristics neurogenic and improve functional recovery in an experimental rat model SCI. ADSCs from rats were cultured purified vitro, followed by genetically modified with gene. Forty-eight adult female Sprague–Dawley randomly assigned three groups: control, ADSCs, Ngn2–ADSCs groups. The hind-limb motor function all was recorded using Basso, Beattie, Bresnahan locomotor rating scale for 8 weeks. Moreover, hematoxylineosin staining immunohistochemistry also performed. After neural induction, positive expression rate NeuN group upon 90 %. Following transplantation, great number found around center injury spinal cord at 1 4 weeks, which improved retention tissue lesion site. differentiated into neurons, indicated markers, Tuj1. Additionally, transplantation upregulated trophic factors (brain-derived neurotrophic factor vascular endothelial growth factor), inhibited glial scar formation, fibrillary acidic protein. Finally, Ngn2–ADSCs-treated animals showed highest among These findings suggest Ngn2-overexpressed promote SCI, local microenvironment injured more efficient way than alone.
