摘要: CD1d-restricted natural killer T cells (NKT cells) possess a wide range of effector and regulatory activities that are related to their ability secrete both T helper 1 (Th1) cell- Th2 cell-type cytokines. We analyzed presentation NKT cell activating α galactosylceramide (αGalCer) analogs give predominantly cytokine responses determine how ligand structure controls the outcome activation. Using monoclonal antibody specific for αGalCer-CD1d complexes visualize quantitate glycolipid presentation, we found cytokine-biasing ligands were characterized by rapid direct loading cell-surface CD1d proteins. Complexes formed association these αGalCer with showed distinctive exclusion from ganglioside-enriched, detergent-resistant plasma membrane microdomains antigen-presenting cells. These findings help explain subtle alterations in can control balance proinflammatory anti-inflammatory
