摘要: This study provides the first immunohistochemical evidence visualizing cholinergic octopus neurons containing choline acetyltransferase (ChAT), synthetic enzyme of acetylcholine. Because antiserum applied here was raised against a recombinant protein encoded by exons 7 and 8 rat gene for ChAT, initially used studies in mammals, to validate antibody specificity counterpart we therefore three methods. Immunoprecipitation using Pansorbin indicated that immunoreactive brain molecules were capable synthesizing Western blot analysis after denatured gel electrophoresis extracts revealed single band at approximately 81 kDa. A slice 81-kDa native (nondenatured) exhibited high ChAT activity. All findings obtained with these methods clearly effectively recognizes ChAT. The use retina, optic lobe, its neighboring peduncle complex detected enzyme-containing neuronal cell bodies only two regions, islands lobe medulla cortical layer posterior olfactory lobule. Immunoreactive fibers probable nerve terminals also found plexiform deep within stroma gland, neuropils lobe. These results provide information on morphology distribution patterns visual system, useful invertebrate model learning memory where as higher vertebrates including plays an important role.
