摘要: Antisuppressor mutations reduce the efficiency of nonsense suppressors. A mutation in gene sin4 Schizosaccharomyces pombe leads to loss 5-(methoxycarbonylmethyl) thiouridine (mcm5s2U) from first anticodon position tRNAs. This resembles phenotype sin3 (Heyer, W. D., Thuriaux, P., Kohli, J., Ebert, Kersten, H., Gehrke, C., Kuo, K. and Agris, P. F. (1984) J. Biol. Chem. 259, 2856-2862), but reside different genes. In vivo 35S-labeled tRNA parental suppressor strain sup3, antisuppressor strains sin4, double mutant has been digested nucleosides analyzed with high performance liquid chromatography methods. The major sulfur-carrying nucleoside wild-type S. is mcm5s2U. It reduced strains. Two other thiolated are also present: 2-thiouridine a unknown structure. Neither was affected by mutations.
