摘要: The screening of antisuppressor mutants the yeast Schizosaccharomyces pombe has been successfully accomplished with high resolution liquid chromatographic methods for analysis tRNA nucleosides. Antisuppressor mutations reduce or abolish function nonsense suppressor-tRNAs other informational suppressors. Nonradioactive 35S-labeled unfractionated from various strains was digested to nucleosides and analyzed by performance chromatography. mutant sin3 lost nucleoside 5-(methoxycarbonylmethyl)-2-thiouridine its in comparison parental strains. In eukaryotes this is found at first position anticodon (wobble position) several isoacceptor tRNAs that preferentially recognize codons ending adenosine. mutation reduces efficiency UGA UAA suppressor tRNASer tRNALeu. genetic cosegregation modification loss, phenotype, a change cell size demonstrated. This indicates single structural gene enzyme causes three different phenotypes.
